PEPTIDE PROTEIN CHEMISTRY FACILITY

Faculty working in Peptide Core

The Peptide and Protein Chemistry Core Facility at the University of Colorado, Anschutz Medical Campus provides state-of-the-art instrumentation for solid-phase peptide synthesis using Boc- and Fmoc-chemistry, peptide purification by rev​ersed-phase HPLC and peptide identification by mass spectrometry (LC-MS) services for academic and industrial clients.

Core Facility personnel will help users select and design the appropriate peptide sequences for their applications including how and where to incorporate fluorescent and biotin labeled probes, incorporation of modified and isotopic labeled amino acids and covalent constraints to stabilize the peptide conformation, etc.

Peptide Synthesis (Solid-phase / solution phase)

Peptides can be synthesized using L- and/or D-amino acids or unusual amino acids. Boc-, Fmoc- or a combination of these synthesis chemistries can be used. 


Peptide Purification (Reversed-phase HPLC)

Our facility only provides synthetic peptides at purities greater than 95% at the milligram to gram scale. All peptides are analyzed for purity by analytical reversed-phase HPLC and characterized by LC-MS (electrospray mass spectrometry).

Peptide Conjugation (polyclonal/monoclonal antibody production)

We can couple synthetic peptides to carrier proteins for production of polyclonal or monoclonal antibodies. We generally couple to Keyhole limpet hemocyanin (KLH) for antibody production and Bovine serum albumin (BSA) for screening antibodies by ELISA.

PEPTIDE SYNTHESIS

Our staff has over 100 years of experience in peptide design and synthesis, and we are happy to discuss your order with you at no charge. Customers will receive a quotation prior to beginning production to assess costs. The costs of synthesis are dependent on peptide length and the incorporated modifications. Some bulk discounts may be available depending on the project.


Minimum charge for synthesis ($250)
0.1 mM scale: $25 per residue
0.25 mM scale: $40 per residue
All peptides are cleaved from the resin, resolubilized in appropriate solvent, and analyzed by reversed phase HPLC and LC-MS
Preparative HPLC purification of synthetic peptides: $250 per peptide (depending on scale of synthesis)
Additional modification of peptides:
Incorporation of phosphorylated Ser or Thr
Incorporation of D-amino acids or other unusual amino acids
Acetylation
Biotin (C-terminal or N-terminal)
